Wake Hard Disk from Sleep?

Out of curiosity, in laptop specifically, if I have a secondary hard disk that I use to store relatively static data, such as photos, that drive will be put to sleep by the OS because it is not being actively used. If a folder on that drive is part of a Syncthing relationship, will syncthing wake that drive from sleep constantly?

On every scan interval Syncthing will list all files and (sub)folders in that folder, which should result in the OS waking up the drive.

If you want the drive to be sleeping as long/often as possible, you should set the rescan interval accordingly and/or use inotify, which should not interfere with the sleeping.
